The recently updated NCode™ Multi-Species miRNA Microarray Kit V2 contains optimized probes targeting all of the known mature miRNAs in mirBase 9.0 for human, mouse, rat, Drosophila, C. elegans, and zebrafish, spotted in triplicate on Corning Glymo epoxide slides (Table 1). In addition to the increased species coverage, the V2 array contains:

• Increased mismatch controls for monitoring hybridization specificity
• Updated NCode™ Positive Control for labeling quality control and easier spot finding
• Dye normalization controls to quickly normalize fluorescent channels

Probe design
Careful consideration must be given when designing probes for interrogating miRNAs, to ensure maximum specificity and sensitivity. The probes on the NCode™ Multi-Species miRNA microarray V2 were designed using a validated probe design algorithm that yields:

• Maximum hybridization intensities for increased sensitivity (Figure 1)
• Maximum specificity for discerning closely related miRNAs (Figures 1 and 2)
• Normalized melting temperatures for uniform hybridization

Contents and Storage:
The NCode™ Multi-Species miRNA Microarray Kit V2 contains five microarrays. Each array is spotted in triplicate on Corning Glymo epoxide slides. The NCode™ Multi-Species miRNA Probe Set is provided lyophilized in a 384-well format. The NCode™ Multi-Species miRNA Microarray Control V2 is a single synthetic RNA sequence provided in solution at 2 pmol/µl. Store microarrays at room temperature. Store the Probe Set and Microarray Control at -20°C. Guaranteed stable for 6 months when properly stored.
