The CD8A, clone 5H10, Pacific Orange™ dye conjugate is a monoclonal mouse antibody that is reactive to the mouse CD8α (Ly-2) antigen. Our Pacific Orange™ dye is optimally excited by the violet laser. Pacific Orange™ and Pacific Blue™ dye conjugates can be simultaneously excited at 405 nm and emit at 551 nm and 455 nm, respectively, facilitating two-color analysis.


Specificity for CD8α
The 5H10 monoclonal antibody (mAb) recognizes the mouse CD8α chain. The α chain of CD8 associates with the CD8β chain to form a CD8α/β heterodimer that is expressed by the majority of thymocytes and by the MHC class I restricted subset of mature T cells. Mouse CD8α can also form a CD8α/β chain homodimer on subsets of CD8 positive T cells. For this reason, antibodies specific for CD8α rather than CD8β are recommended for a rigorous delineation of CD8 positive T cells. The 5H10 mAb has been used in other labs for flow cytometry and inhibition of T-cell responses to IL-21.
