The CD8α, clone 5H10, Pacific Blue™ conjugate is a monoclonal rat antibody that recognizes the expressed product of the Cd8α gene and is specific to mouse CD8α. Our Pacific Blue™ dye is optimally excited by the violet laser, and conjugates of this dye are strongly fluorescent even at neutral pH. Pacific Blue™ and Pacific Orange™ dye conjugates can be simultaneously excited at 405 nm and emit at 455 nm and 551 nm, respectively, facilitating two-color analysis.


Specificity for CD8A
The 5H10 monoclonal antibody (mAb) recognizes the mouse CD8α chain. The α chain of CD8 associates with the CD8β chain to form a CD8α/β heterodimer that is expressed by the majority of thymocytes and by the MHC class I restricted subset of mature T cells. Mouse CD8α can also form a CD8α/α chain homodimer on subsets of CD8 positive T cells. For this reason, antibodies specific for CD8α rather than CD8β are recommended for a rigorous delineation of CD8 positive T cells. The 5H10 mAb has been used in other labs for flow cytometry and inhibition of T-cell responses to IL-21.
