The AKT Pathway Phospho 7-Plex Panel for the Luminex® platform is specifically designed for quantifying the phosphorylated proteins IR[pYpY1162/1163], IGF-1R[pYpY1135/1136], IRS-1[pS312], Akt[pS473], PRAS40[pT246], p70S6K[pTpS421/424], and GSK-3β[pS9] in cell lysates and tissue homogenates (Figure 1). The antibodies used in this assay are human, mouse and rat cross-reactive. This kit has not been tested for multiplexing with other markers. Should user elect to multiplex this kit with other Luminex kits, the assay conditions should be determined empirically for each specific application. By measuring 7 analytes simultaneously, the Luminex® assay panel helps provide more data from each sample, saving both money and time compared to more traditional systems (such as ELISA) used in research. This assay panel is suitable for use with the Luminex® 100™/200™ and FLEXMAP 3D® systems.

Luminex® xMAP® Technology-Effective and Proven Analytical Tool
The AKT Pathway Phospho 7-Plex Panel is based on xMAP® technology. The use of a suspension bead-based technology enables the multiplexing capabilities of the Luminex® assays. Polystyrene microspheres are internally dyed with red and infrared fluorophores of differing intensities. Each bead is given a unique number, or bead region, allowing differentiation of one bead from another. Beads covalently bound to different antibodies can be mixed in the same assay, utilizing a 96-well microplate format. At the completion of the sandwich immunoassay, beads can be read using the Luminex® 100™/200™ and FLEXMAP 3D® detection systems, instruments based on flow cytometry, two lasers, real-time digital signal processing, and xPONENT® software to distinguish bead color (analyte) and assay signal strength (PE) fluorescence intensity.
