The SilverQuest™ Silver Staining Kit provides maximal silver staining sensitivity with minimal protein modification for optimal mass spectrometry results. The mass spectrometry problems associated with traditional silver stains are eliminated with the SilverQuest™ Kit because the reagents used do not modify protein side chains and do not interfere with trypsin digestion. In less than 90 minutes, 0.3 ng of protein can be detected (Figure 1). For added speed, an optimized microwave procedure can be completed in 30 minutes without compromising sensitivity.
Mass Spectrometry-Compatible ReagentsThe SilverQuest™ Silver Staining Kit has been specifically designed to make protein detection compatible with mass spectrometry analysis. The SilverQuest™ reagents include:
- A glutaraldehyde- and formaldehyde-free sensitizer that does not modify lysine and arginine side-chain residues, allowing for efficient in-gel trypsin digestion and optimal peptide extraction from the gel (1)
- Destaining solutions that effectively remove silver from protein bands in polyacrylamide gels (2), which results in improved trypsin digestion efficiency and better protein coverage during mass spectrometry analysis (Table 1) In addition, solutions are color-tinted to simplify staining.
