Easy-to-Run Sandwich ELISA
The Human Aβ42 kit is a solid phase sandwich Enzyme Linked-Immuno-Sorbent Assay (ELISA). A monoclonal antibody specific for the NH2-terminus region of human Aβ has been coated onto the wells of the microtiter strips provided. During the first incubation, standards, controls, and unknown samples are pipetted into the wells and co-incubated with a rabbit antibody specific for the COOH-terminus of the 1-42 Aβ sequence. Bound rabbit antibody is detected by the use of a horseradish peroxidase-labeled anti-rabbit antibody. After washing, horseradish peroxidase-labeled anti-rabbit antibody (enzyme) is added. After a second incubation and washing to remove all unbound enzyme, a substrate solution is added, which is acted upon by the bound enzyme to produce color. The intensity of this colored product is directly proportional to the concentration of human Aβ42 present in the original specimen. Samples are quantitated by the standard curve generated from the recombinant Aβ42 included with the kit (Figure 1).

Results typically in one day – Rapid 4-hour Incubation Protocol
The kit comes with the capture antibody pre-coated to the wells, eliminating the need for you to coat the plate. No overnight incubation needed. The incubation steps can be performed in 4 hours.

Large Dynamic Range – 7.1-1000 pg⁄ml
The large dynamic range of the assay minimizes the need to guess how much dilution is needed to get your sample to fall within the linear assay range (Figure 2).

Economical Removable Wells
The pre-coated wells come as removable 8-well strips allowing you to tailor the assay size to your needs. Two standard vials per plate are provided to give you the option of reconstituting a new vial should you choose to not run the entire plate at one time.
