The PureLink™ miRNA Isolation Kit is uniquely designed to purify cellular small (< 200 nt) RNA molecules, including tRNA, 5S rRNA, and 5.8S rRNA, and regulatory RNA molecules such as microRNA (miRNA) and short interfering RNA (siRNA) (Figures 1 and 2). Purified RNA is suitable for northern blot analysis and microarray analysis.
The kit utilizes a silica-based two-column system to enrich small RNA from various sample sources. From sample lysate, RNAs greater than ~200 nt are bound to the first column. The flow-through, enriched with small RNA molecules, is mixed with a higher concentration of ethanol to increase the binding affinity of small RNA to the glass fiber membranes of the second column. Bound small RNA molecules are then eluted, ready-for-use, and free of large RNA molecules such as 28S rRNA, 18S rRNA, and mRNA that can inhibit expression analysis of small RNA molecules.
