Purification of viral RNA or DNA from cell-free fluids has traditionally been performed using silica plates or silica magnetic-bead separation methods, resulting in eluted, purified nucleic acids in large volumes. The ChargeSwitch® EasyPlex™ Kit eliminates RNA or DNA elution, allowing for complete use of the nucleic acids from 30 µl serum or plasma samples for enzymatic applications including qPCR and qRT-PCR.

The simple process (Figure 1) involves minimal handling, which allows for easy automation and reduces the possibility of false reports related to human error. In addition, the ChargeSwitch® system does not use ethanol for washing RNA or DNA, which means there is less of a chance of PCR inhibition or reduced nucleic acid recovery from inefficient ethanol removal.

Sensitive viral quantitation of serum and plasma using real-time PCR and RT-PCR

The ChargeSwitch® EasyPlex™ system allows for extraction of both viral RNA or DNA for viral analysis protocols ranging from detection to genotyping. Purity of viral RNA or DNA is a critical factor in viral detection analysis since real-time PCR is sensitive to even minute impurities in the sample. To measure both sensitivity and purity of viral RNA and DNA processed using the ChargeSwitch® EasyPlex™ Viral system, we analyzed the recovery of viral nucleic acids from only 10 µl of serum or plasma, using real-time PCR.

Purification using ChargeSwitch® EasyPlex™ is both sensitive and linear down to the limit of 100 RNA virus particles and 1,000 DNA virus particles using the EasyPlex™ Viral system. No difference was observed for plasma versus serum, although plasma is a more proteinacous sample type (Figures 2 and 3). The data indicate that RNA is of high quality and contaminants in the sample are completely removed, as even trace impurities would have impacted the RT-PCR results.
