The human ERG related gene (Kv11.1, KCNH2) encodes a voltage gated potassium channel known to repolarize cardiac muscle following an action potential. Blockage or perturbation of hERG activity in vivo leads to well described cardiac arrhythmias, and rigorous hERG screening and safety testing is required of all new drugs in preclinical development.
To enable the study of hERG in a variety of mammalian cell types, we offer the full-length human clone of Kv11.1 packaged into a baculovirus gene delivery/expression system. The kit includes a virus stock solution and an enhancer for increased expression. The non-cytopathic and safe BacMam delivery and expression technology enables transient expression in a wide range of cell types, which for certain target classes has been shown to provide results superior to those obtained by using stable cell lines. U-2 OS cells (ATCC HTB-96) highly recommended as a starting point due to the excellent transduction efficiency (>90%).
BacMam-hERG Potassium Channel reagent was designed for use in conjunction with the novel FluxOR™ Thallium Detection Kit (F10016 and F10017) that enables functional screening and discovery of drug blocking activity at hERG. The FluxOR™ Thallium detection kit measures the activity of hERG and other potassium permeant channels with the report of thallium influx by a proprietary fluorescent dye and gives IC50 values comparable to those obtained by other methods.
Cell types successfully transduced with BacMam-hERG and assayed include U-2 OS (highly recommended), HEK 293, tSA-201, CHO, HUVEC and HeLa cells.
Compared to other viral and non-viral gene delivery methods, BacMam technology offers the following advantages:
