This episomally derived human iPSC line has no interference in the cells from the reprogramming genes. Viruses used for reprogramming, such as lentivirus and retrovirus, contain transgenes that can integrate into the host genome, potentially disrupting the genome or causing unpredictable results. This integration free iPSC line provides researchers with a control or baseline from which to compare and study a multitude of aspects of stem cell research.
Characterized and Validated
Whole genome expression and epigenetic profiling analyses demonstrated that the Gibco® Episomal hiPSC Line were molecularly indistinguishable from human embryonic stem cell lines. This episomal hiPSC line has a normal karyotype and expresses endogenouspluripotent markers including Oct4, Sox2 and Nanog by RT-PCR and Oct4, SSEA4, TRA-1-60 and TRA-1-81 by immunocytochemistry (ICC). Further, the line has been shown to differentiate robustly to vascular, hematopoietic, neural and cardiac lineages. The line has been shown to be fully transgene free.
