TSC2 (Tuberous Sclerosis 2 Protein), has been implicated as a tumor suppressor, and mutations in TSC2 lead to constitutive activation of the RAS oncogene RAP1A in tumors. TSC2 complexed with TSC1 can act as a GTPase-activating protein (GAP) for the small GTPase RHEB, which is an activator of the protein kinase activity of mTORC. It also can weakly stimulate the GTPase activity of the Ras-related proteins RAP1A and RAB5 in vitro.
