Specific for Rb [pT821]
Rb, also known as Retinoblastoma, is a tumor suppressor protein that belongs to the Pocket protein family. It is a nuclear protein that is expressed in retinal cells. Rb prevents the replication of damaged DNA by preventing its progression along the cell cycle through G1 to S. The phosphorylation of Rb at threonine 821 is catalyzed by cdk2 complexes such as cyclin E/cdk2 and cyclin A/cdk2. Active (dephosphorylated) Rb binds and inactivates the cellular transcription factor E2F1, which is required for cell cycle progression. Proteins containing a consensus LXCXE sequence, such as adenovirus E1a and HPV E7, can bind and inhibit Rb. In humans, the Rb protein is encoded by the RB1 gene located on the q arm of chromosome 13.
