Specific for Tau [pS396]
Tau protein, also known as microtubule associated protein (MAPT), plays an important role in nucleation and stabilization of microtubules by associating with tubulin subunits. In humans, Tau is predominantly expressed in brain and is responsible for neurodegeneration in case of malfunction due to misfolding, hyperphosphorylation, and self-assembly into tangles. Six isoforms of Tau with varying degrees of affinity to tubulin exist, wherein the C-terminus of Tau (positively charged) associates with microtubule (negatively charged). Numerous serine/threonine kinases, including GSK-3β, protein kinase A (PKA), cyclin-dependent kinase 5 (cdk5), and casein kinase II (CK2) phosphorylate Tau. Serine 396 is phosphorylated by GSK-3β and cdk5 in vitro and in vivo. The gene encoding Tau protein is located on chromosome 17q21, containing 16 exons.
