p38 Mitogen-activated protein kinase, also known as SAPK2 (Stress-Activated Protein Kinase 2), belongs to the mitogen-activated protein kinase (MAPK) family, which is responsive to stress stimuli. The mammalian p38 MAPK family is activated by cellular stress including UV irradiation, heat shock, high osmotic stress, lipopolysaccharide, protein synthesis inhibitors, proinflammatory cytokines (such as IL-1 and TNF-alpha), and certain mitogens, which may lead to autoimmunity. There are four p38 kinases - α, β, γ and δ. p38α is well characterized and is expressed in most cell types. The p38 MAPK subfamily plays important roles in cytokine production and the stress response. Additional functions of p38 MAPKs are involved with the inhibition of cell cycle progression, developmental processes, and the differentiation of several vertebrate cell types. An important target of the p38 MAPK is the tumor suppressor protein p53, whose activation is positively regulated by p38. MKK3, MKK6, and SEK phosphorylate p38 MAPK at threonine 180 and tyrosine 182. Phosphorylation of these residues activates p38 MAPK which in turn activates and phosphorylates ATF1, ATF2, and ATF6.
