Rb (Retinoblastoma) is a tumor suppressor protein, and belongs to the Pocket protein family. It is a nuclear protein that is widely expressed in retinal cells. pRb prevents the replication of damaged DNA by preventing its progression along the cell cycle through G1 to S. Rb is regulated by phosphorylation. Active (dephosphorylated) pRb binds and inactivates the cellular transcription factor E2F1, the function of which is required for cell cycle progression. In humans, the protein is encoded by the RB1 gene located on the q arm of chromosome 13. Aurora B directly phosphorylates Rb at S-780 both in vitro and in vivo, and this interaction plays a critical role in regulating the post-mitotic checkpoint to prevent endoreduplication (redoubling of chromosomes) after an aberrant mitosis.
