Histone H3 is a core component of the nucleosome, and plays a central role in transcriptional regulation, DNA repair, DNA replication and chromosomal stability. Histone H3 undergoes many post-translational modifications compared to other histones, such as phosphorylation, acetylation, multiple methylation, ubiquitination and ADP-ribosylation. Reversible acetylation occurs on N-terminal lysine residues – acetylation removes positive charges from the histones, thereby weakening their interaction with the phosphate backbone of DNA, thus transforming chromatin into a more relaxed structure that is associated with greater levels of gene transcription. Acetylation of histone H3 at Lysine 9 (H3K9Ac) is one of the most well-known epigenetic markers enriched in the promoter regions of activated genes.
