Vertebrates produce four major glucose-phosphorylating isoenzymes, designated hexokinase I-IV. Hexokinase is involved in the first step of the glycolytic pahway as well as other pathways and catalyzes the phosphorylation of glucose to produce glucose-6-phosphate for entry to the glycolytic pathway. Hexokinase is an allosteric enzyme and is inhibited by its product GLC-6-P. The predominant isoform is hexokinase 2, which gets expressed in insulin-responsive tissues such as skeletal muscle. The expression of the enzyme is insulin dependent. HK2 can be found in the outer mitochondrial membrane. The enzymes catalytic activity is associated with the C-terminus while its regulatory function is associated with the N-terminus.
