TRAF3, also known as TNF receptor-associated factor 3, is an adapter protein and signal transducer that links multiple members of the tumor necrosis receptor family to different signaling pathways. This is done by association with the receptor cytoplasmic domain and various kinases. It also regulates innate immune system by modulating signaling via Toll-like receptors. Overexpression of TRAF3 does not induce activation of the commonly known TRAF-inducible signaling pathways, NF-κB and JNK. In contrast, it acts as a negative regulator of the NF-κB pathway. Furthermore, TRAF3 is a positive regulator of type I IFN production. These results indicate that TRAF3 is a critical regulator of both innate and adaptive immune responses. In addition to being a major modulator of immune responses, TRAF3 regulates B cell homeostasis in secondary lymphoid organs.
