The sensitive and scalable protocol consist of only a few steps of pipetting and magnetic separation, with all operations performed in a single tube. Larger sample volumes can be used to enhance detection sensitivity, while the target can be separated, washed, and concentrated into one or more smaller elution volumes for different molecular downstream applications.
A wide variety of starting samples can be applied (e.g. total RNA or crude extracts of animal tissues, plants or cells). A variety of clinical samples can be used without inhibiting amplification. Elution of the isolated mRNA is not required, since the beads do not interfere with downstream enzymatic reactions. The bead-bound oligo-dT can also serve as a primer for reverse transcription and PCR.

Binding capacity:
Up to 2 µg poly A+ RNA can be isolated per 200 µl (1 mg) of beads, depending on the tissue or cell type and the expression level of the mRNA. A typical mammalian cell contains about 10-30 pg of RNA of which 1 - 5% is mRNA. The total capacity per ml of beads is approx. 10 µg mRNA. The amount of coupled oligo-dT25 probe is70 pmol⁄mg beads.
