Ion TargetSeq™ Exome kits provide a solution-phase DNA probe capture technology for targeted resequencing applications that enables selective, specific, and economical enrichment of exons and other important regions within the human genome. The capture protocol and capture probe content have been thoroughly optimized to give superior performance on both Ion Proton™ and PGM™ sequencing instruments. Ion TargetSeq™ Exome kits employ a capture protocol that requires as little as 500 ng of individual or pooled barcoded libraries, helping to conserve precious clinical samples. The Ion TargetSeq™ protocol allows users to leverage barcoded fragment libraries to provide fast, efficient, and economical sample enrichment prior to sequencing.
Comprehensive Capture Probe Content
The genomic regions that encompass the capture probe content were identified using the latest versions of human gene expression, genome annotation, and cancer databases including RefSeq, CCDS, VEGA, GenCode, Ensembl®, and COSMIC. Specifically, the targeted regions consist of 39.5 Mb of exons, 1472 miRNA genes, >44,000 miRNA binding sites in 3’-UTRs, various tRNAs, snoRNAs, lncRNAs, and >97% of the genes variants in the COSMIC database.
Everything Needed for Target Enrichment
Ion TargetSeq™ Exome kits are conveniently packaged with all the reagents needed to perform target enrichment, including all hybridization and wash buffers, Human Cot-1 DNA® Fluorometric QC, Dynabeads® M-270 Streptavidin magnetic beads, and blocker oligos for standard fragment library adapters, paired-end library adapters, and Ion Xpress™ barcoded library adapters (BCs 1-16). These enrichment kits provide highly specific and reproducible enrichment of 4 to 192 samples with unparalleled coverage metrics (e.g., >90% uniformity of coverage, >75% of reads on target with bases covered at 20x depth).
