Rare Event Detection in Dramatically Reduced Acquisition Times
The Attune® Acoustic Focusing Cytometer is designed to collect up to 20 million events per run. The Attune® Acoustic Focusing Cytometer uses an innovative technology that exploits the physical difference between cells or particles relative to the background medium, allowing adjustable collection rates ranging from 25 µL/min to 1000 µL/min, which is nearly an order of magnitude higher than a conventional flow cytometer. The time required to acquire 1 million events using a hydrodynamic flow cytometer with a HIGH (120 µL/min) flow rate, which is the least sensitive mode and Attune® cytometer at different collection rate is listed in the table below:

The Attune® Acoustic cytometer achieves sample throughput at rates over 10 times faster than other cytometers—up to 1,000 µL per minute, enabling rapid detection of rare events with a reliable measure of accuracy without aborting any data.
High Collection Rates Without Compromising CVs
Traditional flow cytometers utilize hydrodynamic focusing that wraps an inner sample core with a outer layer of sheath fluid to align the cells. As the sample introduction increases, the sample core stream widens and cells become less focused resulting in increased variability of signal. In the Attune® Acoustic Focusing Cytometer, the cell alignment is maintained in the precise center of the sample stream even as the sample stream widens with the increased rate of sample introduction. This tight alignment results in:
• Improved data quality by minimizing variability as cells pass through laser
• Reduced CVs, leading to better resolution
• Increased sensitivity achieved with variable flow rates that allow longer laser dwell times
Simplify Sample Preparation—Maximize Efficiency
Traditional flow cytometers rely on a large volume of fluid to focus cells, limiting the amount of sample that can be used. This often means that cells must be prepared or concentrated prior to analysis on those types of instruments. The Attune® Acoustic Focusing Cytometer offers flexibility in the sample concentration that can be analyzed. Acoustic focusing enables:

• Effective background fluorescence reduction by a simple dilution step, eliminating the need to wash fluorescence samples before analysis

Directly Measure Absolute Counts Without the Use of Beads
The Attune® Acoustic Focusing Cytometer uses accurate syringe pumps to measure the exact volume of sample analyzed during data collection. Using this information, the Attune® cytometer is able to directly measure absolute cell numbers without the need for beads, which minimizes the amount of sample required and helps reduce the costs associated with the assay.
How Acoustic Focusing Works
The Attune® Acoustic Focusing Cytometer is the first cytometer that uses ultrasonic waves (over 2 MHz, similar to those used in medical imaging) to position cells into a single, focused line along the central axis of a capillary.
Acoustic focusing is largely independent of the sample input rate, enabling cells in the Attune® Acoustic Focusing Cytometer to be tightly focused at the point of laser interrogation, regardless of the sample-to-sheath ratio. This, in turn, allows cell velocity to be slowed so that more photons are collected, giving you high-precision analysis at unprecedented volumetric sample throughput.
